DESCRIPTION

Recombinant Proteinase K (produced in Pichia Pastoris) (E1402) from Titirachium album fungus is a serine protease with a wide substrate specificity, even in the presence of detergents.
It has a specific activity of U/mg protein. 1 U of enzyme is defined as the activity generated by peptides and amino acids. Folin-positive equivalents at 1 µmol of tyrosine per minute at 7.5 pH at 37°C.
Recommended working concentration is 50-100 µg/ml for protein digestion and enzymatic inactivation and of 2 mg/ml for tissue treatment. It is recommended to prepare a stock solution of 20 mg/ml with the Proteinase K Buffer (B0221) especially formulated by INBIO HIGHWAY.
Its optimal activity is at 50-55°C. It is stable in a broad range of pH: 4.0-12.5, being optimal at 7.0 to 8.0 ph.
Enzymatic inactivation is produced by heating at 95°C for 10 minutes.
DNAse activity: there is no activity with lambda DNA after 6 hrs of incubation at 37ºC.
RNAse activity: there is no ribonuclease activity after 16 hrs of incubation at 25ºC.

CONSERVATION

Lyophilized proteinase K is transported at room temperature. Once received, keep at 4°C and, after resuspension, store at -20°C.
